Molecular cloning of the gene for the soybean trypsin inhibitor(Kunitz type)

Abstract
To understand the structure and regulation mechanism of the proteinase inhibitor gene, genomic clone for the soybean trypsin inhibitor has been isolated. It was isolated by plaque hybridization of a soybean genomic library with the cDNA clone of SKTI-a as a molecular probe. The genomic library was constructed into EMBL 3 vector with soybean genomic DNA digested with Sau 3AI. Nucieotide sequencing was carried out by Exo III unidirectional digestion in combination with Sanger method. The insert size of the genomic clone G21/RI was about 2.7kb. Genomic Southern blot analysis demonstrated that the soybean genome contains at least three genes coding for the SKTI. Nucleotide sequencing of the clone shows one exon of 615bp which encodes 205 amino acids of the SKTI. The clone includes 1,500bp of upstream and 564bp of downstream sequences. The upstream sequences include the TATA box, seed protein specific sequence $quot;CATGCAT$quot; and the downstream sequences include the five polyadenylation signal sequence, $quot;AATAAA$quot;. Transcriptional regulatory function of those elements will be discussed.
